LDS sample loading buffer, non reducing (4X) is used to prepare protein samples for denaturing polyacrylamide gel electrophoresis using Bis-Tris and Laemmli gels.
- Pre-mixed for convenience
- Requires cold storage
- 4X concentration allows more volume for protein per lane
- Bring to room temperature prior to use
Mixing LDS sample loading buffer with protein samples only denatures the proteins without reducing them. Reducing agents such as DTT or 2-mercaptoethanol can be added to the buffer for protein sample preparation under reducing conditions. Glycerol is included in the loading buffer as a density agent to ensure the samples sink into the sample wells, while Coomassie® serves as a tracking dye to monitor the progression of electrophoresis.
